How does circRNA sequencing work?

Circular RNA (circRNA) is a highly stable molecule of ncRNA, in form of a covalently closed loop that lacks the 5’end caps and the 3’ poly(A) tails. The circular structure grants to circRNAs resistance against exonuclease digestion, a characteristic that can be exploited in library construction.

    Novogene circRNA Sequencing Project Workflow

    The workflow begins with the evaluation of sample quality. After ribosomal RNA (rRNA) depletion, samples are digested by RNase R to remove linear transcripts. Strand-specific library (also known as stranded library, directional library) is prepared, and the sequencing is performed using a paired-end 150bp strategy. Quality assessment of the samples, library, and data results performs in each step to guarantee the delivery results. Bioinformatics pipelines can analyze tailored to multiple needs, providing high-quality, publication-ready results.
